By default users cannot simply reply to a thread via E-mail.
You will have to setup a mailing list and sync messages to the forum.
For details, see Mailing List Manager.

Ok, so I'm looking to find out about the email capabilities of FUDforum.

What I need is to allow users to reply in a thread via email. Obviously the forum notifies you when something happens in a thread or forum, you're watching - so that's a start!
